I actually think that they may have done a blood pact kinda thing. You know, the one where both make a cut on their thumbs and them proceed to "exchange" blood.
It's what I get from that line anyway.

Like, Mundus and Sparda made a promise to watch out for each other and Sparda helped Mundus rise to power as demon king. They had a lot of trust for each other, and going through all that together I guess made them trust each other like they were brothers, but not actual related brothers.

I wonder if they were part of a brotherhood of some kind, and the blood pact was part of the initiation. Thus making way for other "blood brothers" seeking revenge or something.

If they were brothers than Vergil would have just said "brothers". Also, if he was their uncle it would have played part with the story so Mundus probably would have called Dante nephew.

I did have an inkling suspicion that Mundus was just jealous, hence the heart-eating. Dante and Mundus were made for each other.
