I guess either your controller malfunctioned,or maybe there are some errors on your DVD.Because when I played DMC3 SE,it was fine.No problems on locking on to enemies.

Make sure you keep R1 held down and don’t let go of it. If your enemy moves too far away Dante can track down another.

Also, and this may seem obvious but I’ll still point it out, if you kill your targeted opponent Dante will lock onto another!
