No. To unlock her on Dante's disk, you beat it on hard. To unlock her on Lucia's, you beat her story on hard.
I like playing as Lucia because she's faster.
Also, did you know Trish was an unlockable playable character?
It's a demonic fish that you fight as Lucia. She has two underwater levels that are exclusive to her and her only.
The Infested Chopper and Bolverk are exclusive to Dante, though. And so are Argosax the Chaos and The Despair Embodied.
She has a fight with Tateobesu which is also exclusive to her.
That's because he's Lucia exclusive.

He has a white, blue, and green lifebar in addition to yellow orange and red. The only boss that comes close to that is Phantom or Arius-Argosax.
