I don't mean to offend, but wasn't this question asked by many other users in different ways? I mean, I know I'm being hypocritical because I've started a thread like this myself... but ok, let's just move on with this one.
Ahem... Well, I would do a world-conquering game with Vergil as a Samurai/Ninja/Mage-hacker type... so yeah, basically DmC2.
But see, it would be kind of like System Shock 2, where you can choose your abilities (Fighter/Stealth/Cyber-magic) in three different classes. And Sparda can be Shodan/Frank Fontaine/Glados -- then as a plot twist, Sparda is actually Arkham, controlling Vergil this whole time.
Ok, so I don't have an original idea in my head, and only succeded in merging the best ideas in PC storytelling with the stupidest ideas in DMC3.
And if you're thinking "moral choice," I say "not necessarily" -- there was a vid in which Yahtzee reviews Alpha Protocol and he describes the protagonist as the "ponciest ponce that ever ponced past a pawn shop" (citation needed).
This isn't a question of, "can Vergil be redeemed" a-la "The Old Republic" -- this is a question of "how far down the ruthless dick scale can you really go?"
Anyway, that was just my suggestion.

one of my favorite episodes from the entire DCAU. still don't get how creeper is such a bad ass (even cooler than deadpool...CHANCEY) and in arkham city he was getting pummeled by random street thugs.
I think it was the Joker "chemicals" that made him agile. Yeah, I don't get it, either. And if you want to go the "Nolan" realistic route, then he has a split personality (with trained agility), triggered only when he feels the need to put on Joker makeup (for justice). Yeah, that doesn't make sense, either.
actually in the new 52, the creeper was a ancient demon who started to possess Jack Ryder so having a demon possesing a human would work similarly.
i would say a member of the order is chosen to posses a demon and Virgil does this sigil allowing the human to control how he can change into the demon and use its power.
I would reintroduce Nero.
He's not my favourite character,but I feel that if they reintroduce him they can actually bring more depth to this character,you know,more backstory,or development,and actually give him his own personality,because I felt like he was a younger copy of Dante,of course,a version with more angry issues,but he still reminded me of him.
And also,being a young character,he has time to develop as a person,and maybe add some more flaws to him and focus on his grow up process.

For new characters, I'd like to see an angel, considering the only one we saw was Eva, and that was only briefly.
As for old characters, I'd want Credo back, maybe to represent the angels that weren't in DmC. Also, I'd want Agnus to return because I have a feeling he could be made more creepy and menacing than he was in DMC4. Maybe also give more information about his experiments.

Honestly with Loopy on this but Credo. And for me personally, it would be Nero.
Why Credo?
Because he looked and was somewhat interesting, I wanted to see a bit more with him because his fight in DMC 4 was fun.
Nero I would like to see his past revealed(Not depending on a novel) and to keep his attitude.

For Nero, I'd use an early concept art of Dante that NT had.
The one with the black shaggy hair and the red shirt with the star on it. Except make his right arm more demonic and vein-like. His clothes will be like new Dante's hooded coat clothes but the coat's hood would be smaller and dark purple. He'll wear what that concept art is wearing, but the shirt would be black, and the pants would stay the same. He'd wear longer combat boots and his left arm wrapped in tape again. His gun will be a long black revolver pistol and his sword be a long machete that's the same size as old Nero's red queen but is reved up from New Nero's demonic power instead of just a revved up engine.
For Dante he'd have his hair to look like Tony Starks from Iron Man 2.
But his clothes will be like the second DMC4 trailer.
Only he'd wear combat boots instead and he'd have fingerless black gloves.

I would probably have the jason stathan esque Dante design be Nero's dad Rodan and they are a father son team with Rodan being a enforcer for a demonic loan shark and Nero helping him out learning the trade as it were.
They had a run in with Dante a year or two before the events of the game so after the events of the game a older Nero enlists Dante to go spring his dad whose captured by someone.
Instant DLC.
He and his dad are cambions with Rodan being a Half breed so Nero keeps his quarter demon status. Rodan fights with a brawler/martial artist style and is a reference/shoutout to Monkey from enslaved.
There is a funny line on TvTropes that Nero would be introduced as a rebelious skater kid so I would stay true to that but his style focuses on his mental abilities with some fisticuffs.

Dante - has the blood of angels, demons, and humans; leader of the "Devil May Cry" special operations task force. His personality is that of DMC1 Dante with a mix of DMC3 Dante.

Sparda - as he is
Vergil - at first like DMC3 Vergil (the first villain); but in the end, Dante offered his help but was imprisoned by the government for his crimes. Dante comes to visit him in prison often. He served his time and now makes amends by joining the spec ops group "Devil May Cry" as Dante's adviser and master tactician and hacker (like DmC Vergil pre-Downfall). He still dislikes firearms. He is also like the Power Rangers Time Force character Eric Myers (who is also portrayed by Vergil's VA Daniel Southworth); lust for power even at the cost of his own teammates, including Dante.
Lady - reintroduced using her real name (Mary). A CIA agent (considering her VA Kari Wahlgren voiced CIA agent Sonia AKA Irene Lew from Ninja Gaiden 2/Sigma 2) but then joins Dante's group as firearms expert.
Trish - reintroduced as Beatrice (to whom Trish was named after); she will be like DMC Trish, a demon made by Mundus in Eva's image. She deceives Dante and Vergil, like Trish did to Dante in DMC1, (EDIT) but after everything, she joins Dante's group.
Lucia - a half-angel huntress who was at first assigned by the millionaire Arius to kill Dante and Vergil. Later betrayed by Arius (as he views her as a mere tool) and now vows revenge against Arius for his betrayal. In the end, she joins Dante's group.
Arius - a popular millionaire; in secret, he serves an unknown cult.
Arkham - Mary's father. A human who is the sorcerer of an unknown cult. Mary hated him for killing her mother right in front of her eyes when she was very young.
Sanctus - leader of The Holy Order (Order of the Sword reintroduced), but is also the leader of the unknown cult.
Agnus - scientist of The Holy Order, and the unknown cult.
Credo - royal knight of The Holy Order; was brainwashed by Sanctus and Arkham now serving as Sanctus' loyal servant who has no regards for his sister Kyrie.
Kyrie - reintroduces not just as a songstress but a dancer too. Her dance techniques also happen to be some battle techniques.
Nero - an individualistic member of The Holy Order, a religious order who eradicates demons. The only knight to wield guns (not just a twin-barreled revolver but a modified Uzi too). His Red Queen, like the rest of the knights, is now a more traditional broadsword. His Devil Bringer arm remains intact. He and Kyrie suspected something dark within The Holy Order and fended off the cult. He and Kyrie quit the order and joined Dante's group after knowing the order is empowered by demons.
Kat - reintroduced as Catherine (also nicknamed Cat). A human psychic who can sense the presence of demons (like Kat in DmC for seeing demons in Limbo). She was at first a normal girl making graffiti on the streets but was found by Vergil (like DmC Vergil did for Kat).when he witnessed young men (who are demons in disguise) molesting her and killed them. Cat then became part of Dante's group due to Vergil's recommendation.
Phineas - like DmC Phineas but instead is human; has wise knowledge. Dante, Vergil and the rest of "Devil May Cry" force refer him as "Professor Phineas" due to his distinct knowledge. He used to be with the cult.
Morison - a weapons dealer for the black market at first, but was apprehended and now deals arms for Devil May Cry.
Enzo Ferino - the human informant for Dante, Vergil and the rest of Devil May Cry.
Lilith - a stripper who is actually a demoness in disguise working for the cult.
Mundus - the king of demons. The demon who is worshiped by the cult. He, Argosax, Bolverk, and Sparda were inseparable friends until Sparda questioned Mundus motives and leaves his side. Bolverk went rogue as well, but still hated Sparda.

And to bring Phineas and the Kitty Kat. Phineas I really liked a a character and well, I'd be ****ed if Kitty Kat didn't make so much as a cameo.
I'd like to seem them all back too. Kat was a good character as far as females in DMC go.
Phineas was pretty mysterious and I think there's a lot of potential for that character to explain backstory considering he knew Sparda and knew about the other nephilim.
