DT Flux is a move which can be done by keeping the Devil Trigger button pressed. You will notice some flashing red dots above the DT runes. It basically unleashes a flux of energy which kills or makes high amounts of damage in the surrounding area. For as long as you keep It pressed, the more damage It will do.

You can let go the button for as many runes as you want to spend for the DT Flux.

^What he said.

Each glowing rune will be spent when you release it.

DT Flux was very useful in the hardest modes, at least for me.
I miss that red/blue explosion killing all mobs in the area...

It's quite useful for DMD and Bloody Palace, and also against bosses when It deals huge amount of damage, especially when used in string attack combos.

I wonder though, how come Capcom omitted this move in DMC4...? They modified it such when one uses DT, a significant damage is dealt but still a fully-charged DT gauge deals more damage in DMC3... I missed this move when I first played DMC4.
