Do you think any of the main cast of characters actually recognize the presence of items found in the game? Red Orbs, Green Orbs, Holy Water, Vitality Stars, the Statue of Time & Space, etc?

I don't believe so. They're only part of the gameplay mechanics, after all.

Nope. Gameplay mechanics. Although they are shown in some cutscenes, so that's proof they exist.

I remember in the manga of DMC3, he had some of the orbs in his shelf.

Outside of Final Fantasy I doubt any video game world does.
